Peptides are chemically sensitive materials, and reconstitution behaviour can be influenced by the exact diluent used. Different diluents may vary in pH, ionic strength, preservative content, buffering capacity, osmolarity, dissolved gases, and trace impurities. Even small differences in these factors can affect how a peptide dissolves and behaves in solution.

From a chemistry perspective, pH is especially important because it can alter the ionisation state of acidic and basic amino acid side chains. This can change the peptide’s net charge, hydrogen bonding, electrostatic interactions, and hydrophobic association. If the local pH environment is unsuitable, the peptide may show cloudiness, floating particles, clumping, gel-like behaviour, aggregation, or precipitation.

Bacteriostatic water, sterile water, saline, acetic acid water, and buffered solvent systems are not chemically identical. Substituting one for another may change the peptide’s solubility profile, particularly for longer sequences, hydrophobic peptides, modified peptides, or peptides with specific salt forms.

For consistency, reproducibility, and batch comparability, only the recommended diluent system should be used unless an alternative solvent is specifically stated in the product documentation.
